However, optimal benefit from these advantages requires a clear understanding of the many options available for running a real-time PCR experiment. Starting with the theory behind real-time PCR, this review discusses the key components of a real-time PCR experiment, including one-step or two-step PCR, absolute versus relative quantitation, mathematical models available for relative quantitation and amplification efficiency calculations, types of normalization or data correction, and detection chemistries. In addition, the many causes of variation as well as methods to calculate intra- and inter-assay variation are addressed.

Introduction
The advent of real-time PCR and real-time reverse transcription PCR (real-time RT-PCR) has dramatically changed the field of measuring gene expression. Real-time PCR is the technique of collecting data throughout the PCR process as it occurs, thus combining amplification and detection into a single step. This is achieved using a variety of different fluorescent chemistries that correlate PCR product concentration to fluorescence intensity (1). Reactions are characterized by the point in time (or PCR cycle) where the target amplification is first detected. This value is usually referred to as cycle threshold (C<sub>t</sub>), the time at which fluorescence intensity is greater than background fluorescence. Consequently, the greater the quantity of target DNA in the starting material, the faster a significant increase in fluorescent signal will appear, yielding a lower C<sub>t</sub> (2).
There are many benefits of using real-time PCR over other methods to quantify gene expression. It can produce quantitative data with an accurate dynamic range of 7 to 8 log orders of magnitude (3) and does not require post-amplification manipulation. Real-time PCR assays are 10,000- to 100,000-fold more sensitive than RNase protection assays (4), 1000-fold more sensitive than dot blot hybridization (5), and can even detect a single copy of a specific transcript (6). In addition, real-time PCR assays can reliably detect gene expression differences as small as 23% between samples (7) and have lower coefficients of variation (cv; SYBR® Green at 14.2%; TaqMan® at 24%) than end point assays such as band densitometry (44.9%) and probe hybridization (45.1%) (8). Real-time PCR can also discriminate between messenger RNAs (mRNAs) with almost identical sequences, requires much less RNA template than other methods of gene expression analysis, and can be relatively high-throughput given the proper equipment. The major disadvantage to real-time PCR is that it requires expensive equipment and reagents. In addition, due to its extremely high sensitivity, sound experimental design and an in-depth understanding of normalization techniques are imperative for accurate conclusions.
The general steps performed during a real-time PCR experiment, from RNA isolation to data analysis, are outlined in (Figure 1). This review is intended to provide an overview of the many facets of real-time PCR, highlighting PCR theory, quantification methods and models, data normalization, types of detection chemistry, and causes of variation.

Telomerase
Because sperm and egg cells are themselves descended from progenitor cells, if there were no mechanism for replacing lost telomere, then all organisms with linier chromosomes (eukaryotes) would be condemned to quick extinction due to Hayflick limits in their reproductive tissues. Clearly, that's not the case. Instead, there are a number of mechanisms in nature that counteract the natural tendency of telomeres to erode over time. Vertebrates, including mammals, use a remarkable enzyme dubbed 'telomerase'. This hybrid molecule, part protein, part RNA, is capable of slowing telomere erosion, halting erosion altogether, or lengthening telomeres beyond those in the parent cell. The genes that produce telomerase are found in every potentially replicating cell in the body, including cells at their Hayflick limits, but the genes that produce telomerase are inactive in the great majority of our cells, for the vast bulk of our lives. Those genes are active across the body only in early fetal development. After that point, telomerase is only found in a few special tissues such as antibody producing immune cells, cells that replenish the gut lining, and sperm producing cells.

Theo hiện tại em nghĩ, ở các tế bào mầm chẳn hạn, trước khi nhân đôi, Telomerase sẽ làm việc trước bằng cách tạo ra một đoạn DNA tận ví dụ là " TTAGGG" dựa trên RNA của nó (vì Telomerase có phần RNA),nhưng vẫn phải có một đoạn là Primer nữa phải không ạ?

In eukaryotes, a core promoter sequence in the DNA must be present for RNA polymerase to initiate transcription. Promoters are regions of DNA that promote transcription - in eukaryotes, they are found at -30, -75, and -90 base pairs upstream from the transcription start site (abbreviated to TSS). Transcription factors are proteins that bind to these promoter sequences and facilitate the binding of RNA Polymerase.
The most characterized type of core promoter in eukaryotes is a short DNA sequence known as a TATA box, found 25-30 base pairs upstream from the TSS.[4] The TATA box, as a core promoter, is the binding site for a transcription factor known as TATA-binding protein (TBP) (which is itself a subunit of another transcription factor, called Transcription Factor II D (TFIID)). After TFIID binds to the TATA box via the TBP, five more transcription factors and RNA polymerase combine around the TATA box in a series of stages to form a preinitiation complex. One transcription factor, Transcription factor II H, has two components with helicase activity and so is involved in the separating of opposing strands of double-stranded DNA to form the initial transcription bubble. However, the preinitiation complex produces only a low transcription rate on its own. Other proteins known as activators and repressors, along with any associated coactivators or corepressors, are responsible for modulating transcription.

Electrical Synapses
Electrical synapses, although rare in vertebrate nervous systems, do exist. In an electrical synapse, or gap junction, the presynaptic and postsynaptic membranes are partially fused. This allows the action potential to cross from the membrane of one neuron to the next without the intervention of a neurotransmitter. Electrical synapses often lack the directional specificity of chemical synapses and may transmit a signal in either direction. Protein denaturation

Protein denaturation involves a change in the protein structure (generally an unfolding) with the loss of activity. It has been described using the small protein, hen egg white lysozyme . Water is critical, not only for the correct folding of proteins but also for the maintenance of this structure. Heat denaturation and loss of biological activity has been linked to the breakup of the 2-D-spanning water network (see above) around the protein (due to increasing hydrogen bond breakage with temperature), which otherwise acts restrictively on protein vibrational dynamics. As proteins denature their structures initially become looser allowing them to take up more water with the water-exposed surface increasing by up to 50% much in the reverse to protein folding. It is a cooperative process (see right). The free energy change on folding or unfolding is due to the combined effects of both protein folding/unfolding and hydration changes. These compensate to such a large extent that the free energy of stability of a typical protein is only 40-90 kJ mol-1 (equivalent to very few hydrogen bonds), whereas the enthalpy change (and temperature times the entropy change) may be greater than ±500 kJ mol-1 different. There are both enthalpic and entropic contributions to this free energy that change with temperature and so give rise to heat denaturation and, in some cases, cold denaturation. Protein unfolding at higher temperatures (heat denaturation) is easily understood but the widespread existence of protein unfolding at low temperatures is surprising, particularly as it is unexpectedly accompanied by a decrease in entropy. Heat denaturation is endothermic (on heating) but cold denaturation is exothermic (on cooling). Cold denaturation occurs only in aqueous solutions due to water's anomalous density behavior and the small size of H2O molecules.
